Is it possible to create a /.well-known/acme-challenge/ folder-routing with the integrated kerio webserver - i mean without installing a extra webserver. to answer requests for a ssl validation.

like: https: //mykerio.com/.well-known/acme-challenge/

There is a /custom folder where you can add your own html files. What exactly is the problem you are trying to solve?

Hi Brian, i generate let's encrypt-certificates on a separate reverse-proxy. for the domain-request it is asking for a /.well-known/acme-challenge directory to the corresponding server to the url. for this i need a url like https: //mykerio.com/.well-known/acme-challenge/ which is reachable from outside. if i can do this on the kerio server, i don't need to create a side-path routing to another server.

You would have to create it inside the /custom folder. Otherwise it will be removed on upgrades. It sounds like this should be a directory set up on your reverse proxy.
If your objective is to set up free SSL, I suggest to look at cloudflare.com. They have a free tier that gives you Comodo SSL and there are no requirements on the backend server. They handle the reverse proxy / SSL for you and they also provide DDoS protection, caching, CDN, DNS, Firewall, analytics...

AFAIK it should still work, but for Let's Encrypt I would use a separate web server with rewrite rules.
